The fluorescence decay functions of individual, specifically labeled t
RNA(Phe) molecules exhibit nonexponential character as a result of con
formational dynamics occurring during the measurement on a single mole
cule. tRNA(Phe) conformational states that interchange much more slowl
y are evidenced by the distribution of lifetimes observed for many ind
ividual molecules. A structural model fur the nonexponential decay ind
icates that the tRNA(Phe)-probe adduct fluctuates between two states,
one of which provides conditions that quench the probe fluorescence.
