A nonlinear optical spectroscopy method is presented, which allows for prec
ise recording of membrane potentials. The signal intensity of second harmon
ic generation (SHG) mediated by hemicyanine dye molecules embedded in the m
embrane of oxidized cholesterol was used as sensitive probe to membrane pot
