Preparative isolation and purification of phenylethanoid glycosides from the extract of faeces of beagle dogs by high-speed countercurrent chromatography

High-speed countercurrent chromatography (HSCCC) was applied to the separat
ion of acteoside and isoacteoside from a crude extract from canine faeces a
fter the dogs were orally administered phenylethanoid glycosides (PhGs). Tw
o-step separation with a quaternary two-phase solvent system composed of et
hyl acetate-n-butanol-ethanol-water (system 1, 35:6:6:50, v/v; system 2, 30
:10:6:50, v/v) was used. HPLC analysis of final CCC fractions revealed that
the two main PhGs showed purity of over 95%. Their chemical structures wer
e identified by (HNMR)-H-1, (CNMR)-C-13, FAB-MS.
