Recent transgenic as well as gene-targeted animal models have greatly
increased our understanding of the molecular mechanisms of normal and
compromised heart function. These studies have raised the possibility
of using somatic gene transfer as a means for improving cardiac functi
on. DNA transfer to a significant portion of the myocardium has thus f
ar been difficult to accomplish. This review describes current efforts
to achieve myocardial gene transfer in several model systems, with pa
rticular emphasis placed on adenovirus-mediated gene delivery, its pos
sibilities, and current limitations. (C) 1997, Elsevier Science Inc.
