Abstract
Major arachidonic acid metabolites released by mouse splenocytes into the incubation medium were identified by chromatography and spectrosco py and by mass spectrometry of derivatives of the metabolites. With or without exogenous arachidonic acid, splenocytes mainly produced 12-hy droxy-5,8,10,14- and 12,20-dihydroxy-5,8,10,14-eicosatetraenoic acids. Both products (the latter predominating) were continuously released b y intact cells without exogenous substrate or modulators. This is the first demonstration that exogenous ganglioside GM(3) and its lactone m odulate the metabolism of arachidonic acid in splenocytes.
