A method is described for the simultaneous determination of protease inhibi
tors Indinavir, Saquinavir, and Ritonavir in human plasma using liquid chro
matography with ultraviolet and electrochemical detection, Sample pre-treat
ment consisted of solid-phase extraction prior to ion-pair reversed phase h
igh performance liquid chromatography with ultraviolet detection at 240 nm
(Saquinavir and Ritonavir) and electrochemical detection at + 750 mV (Indin
avir and Saquinavir). The method was validated with respect to specificity,
linearity, limits of detection and quantification, precision, and accuracy
. The proposed LC assay was utilized to analyze plasma samples from patient
s treated with HIV-protease inhibitors.
