Abstract
Procedures to generate usable microsatellite marker sequences should be opt imized for cost-effectiveness in each species. For hop, we have used a cock tail of several restriction enzymes to digest the genomic DNA. This is foll owed by capture of microsatellite-containing sequences with long microsatel lite probes attached to a membrane. The enrichment level for GA and GT libr aries was 37% and 35%, respectively, and 100% of the clones contained micro satellite sequences. Libraries can be generated from genomic DNA in approxi mately 10 d.
