A protocol for in vitro shoot regeneration from cotyledon explants of Citru
llus lanatus (Thunb.) Matsum. & Nakai cv. Sugar Baby is described. The coty
ledons excised from 7-d-old aseptic seedlings showed the highest percentage
of shoots on Murashige and Skoog (MS) + N-6-benzyladenine (BA; 3.0 muM) N-6-[2-isopentenyl] adenine (2iP; 3.0 muM) and MS + BA (3.0 muM) + indole-3
-acetic acid (1AA; 3.0 muM). Whereas the latter medium induced shoot regene
ration after the callusing of the explant, the former stimulated direct sho
ot formation. The regenerated shoots were rooted and the resulting plants w
ere established in earthen pots with 55% success.
