Abstract
Mutants with symbiotic activity were obtained by random Tn5-Mob mutage nesis of Rhizobium meliloti 41. The place of the Tn5 insertion was loc alized on a cryptic middle-sized plasmid pRme41a. Mobilization of the labelled plasmid pRme41a::Tn5-Mob into R. meliloti 114, which did not contain such a plasmid led to a complete loss of the nodulation abilit y of the recipient strain. The Nod(-) phenotype was a result of a targ e deletion in the symbiotic (pSym) plasmid of R. meliloti 114.
