The purpose of tl-Lis study was to develop an efficient protocol for plant
regeneration from cotyledons of Cassia obtusifolia in order to improve the
plant by means of biotechnology. The fragments of cotyledons from aseptic s
eedlings of C. obtusifolia as explants were cultured in 6 different media,
MS basic elements were supplemented with different plant regulators, and ca
lli were developed at a rate of 90-99% of the inoculated explants within 15
days. On MSB regeneration medium, containing MS minerals, B5 organic compo
nents, and 0. 15 mg L-1 NAA, and 0.5 mg L-1 each of BA, KIN, and ZT, shoots
were regenerated from 30% of the calli derived from cotyledon explants. Ro
ots were induced with a frequency of 96% on half-strength MS medium plus 0.
2 mg L-1 IBA in 20 days. Complete plants regenerated from cotyledon explant
s were obtained and transferred to soil pots with high survival ratios.
