Aflatoxin B-1 (AfB(1)), present in fungus infested crops is highly carcinog
enic and is measured by immunoassays. I-125 labeled aflatoxin B-1 is a key
reagent for development of radioimmunoassay (RIA) which exhibits less inter
ference and better sensitivity than other immunoassays. Since AfB(1) lacks
suitable functional groups for radiolabeling, an oxime derivative of AfB(1)
was synthesised and evaluated by UV-spectrophotometry and H-1 NMR spectros
copy. I-125-histamine was conjugated to AfB(1) oxime by mixed anhydride met
hod and purified by solvent extraction followed by TLC. The tracer obtained
was immunoreactive, stable as ethanolic solution and could be used in RIA.
