The recently developed whole-column imaged capillary isoelectric focusing f
ocusing is an advanced technique for the pI determination of proteins. In t
his study, we initially applied this technique to determine the pI of peanu
t allergens and its antibody, rabbit IgG. Two main proteins, Ara h I and Ar
a h II, were found in the peanut allergen solution. Their pI values were de
termined to be about 4.5-4.7 and 5.1-5.6, respectively. The value of pI for
the antibody rabbit IgG was 6.1-6.5. Second, the immunoreaction selectivit
y of the two main peanut allergenic proteins with antibody rabbit IgG was s
tudied. The CIEF results show that the peanut allergen Ara h II has a large
r affinity for binding to the antibody rabbit IgG than Ara h I. (C) 2001 Jo
