Melatonin has been suggested to play an important role in the pathogenesis
of gastric mucosal lesions. The aim of this study was to examine the in viv
o effect of melatonin on indomethacin induced gastric lesions in rats. The
activity of secretory phospholipase A(2) (sPLA(2)) in relation to the gastr
ic lesion length was also studied. Our results show that indomethacin induc
ed gastric lesions were significantly decreased after melatonin pretreatmen
t (1, 5 and 10 mg.kg(-1) intraperitoneally). Effect of melatonin was relate
d to the PLA(2) activity. Melatonin significantly decreased indomethacin st
imulated PLA(2) activity (p < 0.01). The results show marked protection of
melatonin against indomethacin-induced gastric lesions. Gastric lesions and
the protective effect of melatonin mag be related to the decrease of sPLA(
2) activity.
