H. Watanabe et al., Monoclonal-based enzyme-linked immunosorbent assay and immunochromatographic rapid assay for salinomycin, ANALYT CHIM, 437(1), 2001, pp. 31-38
Salinomycin is one of the most widely used coccidiostats in the Japanese po
ultry industry. A simple and reliable analytical method for this drug is re
quired. The current determination by bioautography is not sensitive and is
labor-intensive. This paper reports an enzyme-linked immunosorbent assay (E
LISA) using monoclonal antibodies and the development of a rapid test kit b
ased on immunochromatography. The ELISA was quantitative in the range 0.05-
25.6 ng ml(-1) (from 0.005 to 2.56 ng) and showed 50% inhibition (IC50) at
0.7 ng ml(-1) (equivalent to 0.07 ng). The detection Limits using the ELISA
were 10 ng g(-1) for plasma, liver and chicken muscle. The detection Limit
using the kit was 50 ng g(-1) for plasma and 300 ng g(-1) for chicken musc
le. (C) 2001 Elsevier Science B.V. All rights reserved.
