A method improved by NEUMANN (1999) for the quantitative analysis of poultr
y haemoglobin following conversion to porphyrin is described. The specimen
materials used were shaken blood and heparinised blood of broiler as well a
s crystalline pigeon haemoglobin. The studies included the examination of t
hese specimen materials for their suitability for this method, the conversi
on of defined haemoglobin solutions (native and crystalline haemoglobins) t
o porphyrins, and their spectrophotometric analysis. The loss of haemoglobi
n in the process of ultrafiltration through a Millipore filter was determin
ed and used to calculate a correction value. The regression equation derive
d on the grounds of the results enabled an unknown haemoglobin concentratio
n to be calculated from a measured extinction.
