A simple method for PCR-based plant clinical diagnosis of bacterial bl
ight of geraniums caused by Xanthomonas campestris pv. pelargonii is d
escribed. The method entails maceration of infected tissues in water o
r 10 mM Tris-HCl, pH 8.0 buffer, followed by treatment of the macerate
with a commercially-available extraction matrix (GeneReleaser(TM)) in
which nucleic acid is released by brief microwave heating. Nucleic ac
id prepared in this manner served directly as template for PCR amplifi
cation with primers targeting a sequence in the genome of the bacteriu
m. Using this protocol, it was possible to quickly identify X. campest
ris pv. pelargonii in infected geraniums, whereas amplification produc
ts were not obtained with nucleic acid preparations from noninfected p
lants, or from plants infected with the bacterial pathogens, Corynebac
terium fascians or Pseudomonas cichorii.
