Abstract
The detergent Triton X-114 was used in the successful extraction of a chloroplast membrane-bound plant enzyme, polyphenol oxidase (PPO, E.C. 1.10.3.1), from tea, Camellia sinensis, on a large (20 mg) scale. Tri ton X-100 and Triton X-114 were compared; Triton X-114 gave 4x more ac tivity. Addition of phenol adsorbent PVPP, protease inhibitor PMSF and snail acetone powder cellulases, and purification by hydrophobic chro matography gave 23 mg PPO, 2.73 Units.mg(-1).
