In the presence of iron and acetate, the rate of increase of Pseudomon
as fluorescens pyoverdine fluorescence was inversely proportional to t
he iron concentration, up to approximately 200 mu g/l. An assay based
on this observation would however require calibration curves to be gen
erated in a matrix similar to that of the sample. Pyoverdine has an af
finity for Fe(ll), suggesting that it may be necessary to convert Fe(I
I) to Fe(III) prior to assay. More detailed work regarding the mechani
sm underlying the change in fluorescence, assay specificity, accuracy
and precision is necessary to establish this method as a new analytica
l technique.
