Two complexes of bovine seminal ribonuclease with dinucleotides, uridy
lyl(2'-5')adenosine (UpA) and 2'-deoxycytidylyl(3'-5')-2'-deoxyadenosi
ne (d(CpA)), were crystallized under unusual conditions involving a li
quid-liquid phase separation. This phenomenon was induced by adding sm
all aliquots of organic cosolutes to highly concentrated ammonium sulf
ate solutions. The liquid-liquid interface acts as a source of nucleat
ion centers for growth of large crystals. Among the cosolutes tested i
n these salt-mediated crystallizations, 2-methyl-2,4-pentanediol was f
ound to be the most efficient.
