Two methods for stereospecific analysis of triacylglycerols are compar
ed. Procedure A, based on stereospecific phosphorylation of sn-1,2 dia
cylglycerols to phosphatidic acids, and procedure B, based on separati
on of the diastereomeric 1,2(2,3)-diacylglycerol methane derivatives b
y high performance liquid chromatography on silica, were applied to ol
ive oil triacyl-sn-glycerols. Statistical evaluation of the results sh
owed good reproducibility, and Student's t-test indicates no statistic
al differences between the two considered procedures, although some sm
all differences were observed and discussed. Fifteen samples of extra-
virgin olive oil, produced in the same region (Umbria, Italy), were an
alyzed with the two considered procedures.
