A method based on HPLC with coulometric array detection for the determ
ination of 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP) in c
igarette smoke condensate has been developed. II consists of a two ste
p purification on a silica cartridge and a preparative reversed phase
column (RP-C-18) followed by a separation on an analytical reversed ph
ase column (RP-Select-B) using acetonitrile/methanol/glacial acetic ac
id/bidist, water 20:10:2:68, v:v:v:v, pH 5.1 as mobile phase. PhIP is
detected in an electrode array detector (eight electrodes) between +58
0 and +700 mV. The average content of PhIP in cigarettes is 20.3+/-1.2
ng/cigarette (n = 6). The recovery tvas 75+/-1.7% (n = 6) and the det
ection limit in the cigarette condensate was found to be 1.3 mu g PhIP
/l.
