A simple and rapid method has been developed for the simultaneous dete
rmination of gizzerosine (GZ) and histamine (Him) in chicken tissues b
y HPLC using an on-column fluorescence derivatization and column-switc
hing technique. GZ and Him in chicken tissues were extracted with perc
hloric acid solution, and the crude extract was purified on a Bakerbon
d COOH(R) cartridge. GZ and Him were derivatized to the respective o-p
hthalaldehyde fluorophore by on-column reaction, followed by column-sw
itching and subsequent chromatographic separation on a reversed-phase
C-18 polymer column with gradient elution. The calibration curves were
rectilinear in the range of 0.02 similar to 1 mu g/mL for GZ, and 0.0
04 similar to 0.2 mu g/mL for Him. The average recoveries were greater
than 97% for both GZ and Him added to chicken gizzard at a level of 1
mu g/g.
