A rapid and highly sensitive method has been developed for the simulta
neous determination of reducing monosaccharides and oligosaccharides b
y normal-phase HPLC with fluorescence detection. The technique of line
ar gradient elution on a TSKgel Amide-80 column was more suitable for
saccharides separation. The post-column reaction was optimized for flu
orometric detection. Under optimum conditions, the detection limits we
re 0.3 similar to 15 ng for the reducing saccharides investigated. The
calibration curves were approximately linear in the range of 2,000 si
milar to 12.5 pmol for glucose, cellotriose, and cellopentaose. The co
efficient of variation for glucose was less than 1%. Application of th
e method for food analysis was successful.
